Species of Epichloë (Ascomycota: Clavicipitaceae), which infect grasses, maintain an obligate symbiotic relationship with flies of the genus Botanophila (Diptera: Anthomyiidae). Sexual reproduction of the fungus usually requires a visit by Botanophila, which serves to ‘pollinate’ the fungus with spermatia of the opposite mating type; the flies in turn deposit their eggs on fungal tissues upon which the larvae feed. For a molecular phylogenetic study, a total of 108 fly larvae were collected from 10 different Epichloë species on various grasses in Europe and North America. Sequence analysis of the mitochondrial cytochrome oxidase gene (COII) detected six distinct Botanophila taxa that were associated with Epichloë. Three were restricted to samples from Europe, two to samples from North America, and one was present on both continents. In North America the common taxon [identified as Botanophila lobata (Collin)] was found on Epichloë hosts of native grasses and on Epichloë typhina (Pers.: Fr.) Tul. and Epichloë festucae Leuchtm., Schardl and M.R. Siegel of the introduced grasses Dactylis glomerata L. and Festuca rubra L., respectively. In a maximum likelihood phylogram, one of the American taxa was most basal, while the other was nested within the European taxa, suggesting that European and American taxa of Epichloë-associated Botanophila spp. may not have separate origins. Analyses of the fly populations further indicated that up to four different Botanophila taxa can be present at a single location. On taxon level there was no specificity of flies for Epichloë host species (except taxon 6 that was found only on Epichloë glyceriae Schardl & Leuchtmann), although several taxa appeared to prefer some hosts over others. Comparison of molecular phylogenetic relationships of Botanophila species with those of the associated Epichloë hosts did not suggest co-evolution of the fungus and the fly. Thus, associations between Botanophila flies and Epichloë hosts may have arisen independently more than once.  相似文献

Mumford Cove, a 48 ha Connecticut embayment on Long Island Sound, has a history of excessive nutrient inputs and corresponding eutrophic conditions with concomitant eelgrass (Zostera marina L.) loss. From 1945 to 1987, a municipal wastewater treatment facility discharged into the cove. In 1987, when the wastewater outfall was diverted to another location, the cove supported a near monoculture of the green algae Ulva lactuca L., covering 74% of the bottom. By 1988, macroalgal areal cover in Mumford Cove had declined to 9%. When we first sampled the cove in 1992, Z. marina and Ruppia maritima L. were present. By 1999, areal distribution of Z. marina and R. maritima had expanded to cover a third of the bottom. Periodic summertime surveys from 2002 through 2004 indicated that Z. marina was present in approximately half of the cove; R. maritima was sparse. Fifteen years after termination of nutrient enrichment, this cove had recovered from 40 years of point source anthropogenic nutrient input, returning from an Ulva-dominated to a Zostera-dominated state.  相似文献

采用GC-MS测定损伤冷蒿VOCs的主要成分,并通过生物测定的方法研究损伤冷蒿VOCs对牧草幼苗根系生长发育的影响。结果表明:损伤冷蒿释放30种VOCs,包含萜烯类、醇类、酯类、醛类和酮类等5类化合物,其中相对含量高于1%的化合物共占VOCs总量的97.37%,桉树脑含量最高(22.43%)。损伤冷蒿释放的VOCs极显著(P0.01)的抑制草木樨、苏丹草、披碱草和冰草幼根的伸长,以及苏丹草侧根与冰草须根的发育,抑制率分别为51.55%、55.34%、37.57%、48.68%、93.04%和25.00%;同时也极显著抑制4种牧草幼根生物量的积累(P0.01)。损伤冷蒿VOCs影响牧草根部中柱组织结构的分化,使草木樨中柱内木质部、韧皮部与形成层不能正常分化;苏丹草、披碱草和冰草的凯氏带发育不完整。由此可见,损伤冷蒿VOCs对牧草根系生长发育具有明显的抑制作用。  相似文献

延安北部丘陵沟壑区退耕还林(草)成效的遥感监测   总被引:2,自引:0,他引:2  
将延安北部丘陵沟壑区的吴起、志丹、安塞、子长、延川、延长和宝塔等7县区做为一个整体,利用GIS综合解译分析SPOTVGT、NOAA/AVHRR、EOS/MODIS、TM和DEM数据,揭示该区域退耕还林(草)生态建设工程所取得的实效。1998-2009年NOAA/AVHRR和EOS/MODIS遥感监测表明,该区域在遥感影像图上凸现,在其北部和西部形成一条明显的与行政区边界相吻合的分界线,表明退耕还林后植被覆盖状况正在逐年改善,而且植被恢复情况明显好于其北部和西部。1999-2007年SPOTVGTNDVI演变表明,该区域NDVI正处于快速上升阶段,线性趋势值为0.0078,与其南部次生林区的NDVI差值在明显减小,与北部粮食种植区的NDVI差值在逐渐加大。退耕前后的TM影像解译结果对比分析说明,延安北部近年来退耕还林(草)的成绩是显著的,生态环境有了明显的改善。与1997年相比,到2007年延安北部有68.37%耕地不再耕种,而大于35°的坡耕地91.45%实现了退耕,25-35°的坡耕地有74.70%退耕,绝大部分高坡度耕地实现了退耕,而较低坡度的坡耕地退耕幅度也很大,达到了50%-70%。退出的耕地主要变为草地、林地和果园。TM影像解译结果表明,林草覆盖度增加非常显著,达到65.3%,增加24.3%,以草的面积增加最为明显。低植被覆盖度的土地面积在大幅度减少,植被覆盖度小于10%的面积减少了83.42%;中植被覆盖度面积在显著增加,到2007年,植被覆盖度在30%-50%的面积最大,占总面积的47.2%,将近一半;高植被覆盖度面积也明显增加,由1997年的6%增加到2007年22%。TM影像与DEM数据叠加分析表明,土壤侵蚀强度总体上降低,强度、极强度和剧烈等级土壤侵蚀的土地面积减少50%以上,但极强度和剧烈的高等级土壤侵蚀面积仍占2007年国土总面积的13.3%,土壤侵蚀状况仍然严重。植被以灌木和草为主,乔木面积较低,森林覆盖率只有22.4%,因此需要继续加强治理。  相似文献

从草鱼Ctenopharyngodon idella肝肾cDNA文库中克隆得到胶原凝集素基因。草鱼胶原凝集素全长cDNA为1128bp,其中5′非编码区229bp,3′非翻译区104bp,最大开放阅读框为795bp,编码264个氨基酸。系统进化分析表明草鱼胶原凝集素与斑马鱼的亲缘关系最近。根据草鱼胶原凝集素序列特征,克隆了包含糖基识别域(CRD)的cDNA,并进行原核表达、纯化获得其重组蛋白PCRD。进行PCRD与6种细菌的凝集和糖抑制实验,结果表明半乳糖、葡萄糖、甘露糖和麦芽糖4种糖都会使PCRD与嗜水气单胞菌的凝集明显下降甚至极大地干扰凝集;麦芽糖使金黄色葡萄球菌的凝集明显下降,而肽聚糖和甘露糖会使凝集受到抑制;此外,PCRD的凝集反应不依赖Ca2+。  相似文献

We assessed vegetation recovery on access roads removed after well abandonment in an active oil‐producing region of northern Great Plains grasslands. We compared extant vegetation on 58 roads, restored 3–22 years previously, to records of species seeded on each and to adjacent, undisturbed prairie, to evaluate main differences between the restored and adjacent community and to explore patterns in the restored plant community over time. The restored plant community was dominated by low richness of seeded non‐native and native grasses and forbs, whereas adjacent prairie had numerous, abundant native graminoids and shrubs and higher richness of native forbs. Cover of seeded species on roads was double that of colonizing species. Disparity in cover of dominant native grasses between the adjacent community and relatively narrow restored roadway suggests that conditions for germination and survival in roadbeds are poor. This is at least partly due to persistence of seeded species. Differences in restored plant composition over time were best explained by changes in species seeded, from non‐natives to natives, and secondarily by successional shifts from ruderal to perennial non‐seeded species. Of the 30 species seeded at least once on these roads, only 10 were commonly used. The long‐term influence of seeding choices in grassland road restorations implies that improvements in these practices will be critical to reversing ecological impacts of roads.  相似文献

Gene order and content differ among homologous regions of closely related genomes. Similarities in the expression profiles of physically adjacent genes suggest that the proper functioning of these genes depends on maintaining a specific position relative to each other. To better understand the results of the interaction of these two genomic forces, convergent, divergent, and tandem gene pairs in rice and sorghum, as well as their homologs in rice, sorghum, maize, and Brachypodium were analyzed. The status of each pair in all four species: whether it was conserved, inverted, rearranged, or missing homologs was determined. We observed that divergent gene pairs had lower rates of conservation than convergent or tandem pairs, but higher rates of rearranged pairs and missing homologs in maize than in any other species. We also discovered species-specific gene pairs in rice and sorghum. In rice, gene pairs with strongly correlated expression levels were conserved significantly more often than those with little or no correlation. We assigned three types of gene pair to one of 14 possible evolutionary history categories to uncover their evolutionary dynamics during the evolution of grass genomes.  相似文献

Aims: To investigate the effect of the forage source and feed particle size (FPS) in ruminant rations on the composition of the ruminal Firmicutes community in vitro. Methods and Results: Three diets, varying in maize silage to grass silage ratio and FPS, were incubated in a rumen simulation system. Microbial samples were taken from the liquid fermenter effluents. Microbial community analysis was performed by 16S rRNA‐based techniques. Clostridia‐specific single‐strand conformation polymorphism profiles revealed changes of the community structure in dependence on both factors tested. The coarse grass silage–containing diets seemed to enhance the occurrence of different Roseburia species. As detected by real‐time quantitative PCR, Ruminococcus albus showed a higher abundance with decreasing FPS. A slightly lower proportion of Bacilli was found with increasing grass silage to maize silage ratio by fluorescence in situ hybridization (FISH). In contrast, a slightly higher proportion of bacterial species belonging to the Clostridium‐clusters XIV a and b was detected by FISH with increasing grass silage contents in the diet. Conclusions: The ruminal Firmicutes community is affected by the choice of the forage source and FPS. Significance and Impact of the Study: This study supplies fundamental knowledge about the response of ruminal microbial communities to changing diets. Moreover, the data suggest a standardization of grinding of feeds for in vitro studies to facilitate the comparison of results of different laboratories.  相似文献

Silicon is a non-essential element for plant growth. Nevertheless, it affects plant stress resistance and in some plants, such as grasses, it may substitute carbon (C) compounds in cell walls, thereby influencing C allocation patterns and biomass production. How variation in silicon supply over a narrow range affects nitrogen (N) and phosphorus (P) uptake by plants has also been investigated in some detail. However, little is known about effects on the stoichiometric relationships between C, N and P when silicon supply varies over a broader range. Here, we assessed the effect of silicon on aboveground biomass production and C:N:P stoichiometry of common reed, Phragmites australis, in a pot experiment in which three widely differing levels of silicon were supplied. Scanning electron microscopy (SEM) showed that elevated silicon supply promoted silica deposition in the epidermis of Phragmites leaves. This resulted in altered N:P ratios, whereas C:N ratios changed only slightly. Plant growth was slightly (but not significantly) enhanced at intermediate silicon supply levels but significantly decreased at high levels. These findings point to the potential of silicon to impact plant growth and elemental stoichiometry and, by extension, to affect biogeochemical cycles in ecosystems dominated by Phragmites and other grasses and sedges.  相似文献

In the northern Great Plains (United States), sites with less than 20% of native species are difficult to restore. We have experimented with a restoration method that shows some promise. It consists of systematically installing simulated small‐scale patches (8.0 m2 in size) over 25% of an old field and then seeding these patches with native species. The working hypothesis is that these patches will generate a constant source of propagules which in time will lead to increases in native species diversity within the surrounding grass matrix. The objective of this paper was to determine whether soil amendments should be used to facilitate the establishment and persistence of native species (primarily forbs) within these patches. We seeded the patches with a mixture of native grass and forb species and applied four soil treatments: P fertilization, C additions, C + P, and a control (no amendments). Results for the first 5 years were as follows: (1) seeded forb richness was mostly unaffected by soil amendments; (2) seeded and nonseeded forb biomass and density were substantially reduced by C additions, whereas they were unaffected or increased under P additions; (3) both seeded and non‐native grass biomass substantially increased with C additions; and (4) there was an inverse relationship between native seeded forbs and non‐native grass biomass. Our conclusions are that: (1) P amendments are a potential tool for enhancing native seeded forb biomass in simulated small‐scale disturbance patches; and (2) C additions, although enhancing seeded grass biomass do not reduce the biomass of non‐native grasses.  相似文献
